Originally built as an opera house in 1927, the lobby still retains its feeling of grandeur and glamour.

After facing difficulties as an opera house, the venue began to produce nonmusicals including a production of Sophocles' Electra which starred Antoinette Perry, whose is the namesake of the Tony Awards.
In 1976, the theatre was sold to two nightclub managers who transformed the venue into the famous celebrity-filled nightclub, to which the bar still pays homage.
Following a construction issue blocking the Henry Miller theatre, the Roundabout Theatre’s production Cabaret moved to Studio 54 in 1998. The Roundabout Theatre company later purchased the theatre in 2003.
